What cam are you using? Seems to me you should be close already with that setup. I wouldn't stroke it, if you're going to go that route, build a different engine and save the 340. Stroker 360 sounds good and a lot of your parts like the Edelbrock heads would swap.
Also, are the heads straight out of the box? A fresh 3 angle valve job and some bowl clean up might gain you some power as well.
Haven't bought street/strip tires in awhile, so can't comment there.

I don't think you'll need to stroke it. Alot of fellas have bolted in a MP 360 create short blcok with zero deck pistons and a RPM package up top. The short block comes with a purple 292/.509 cam. Everything else the same as you listed, the best I seen someone run is 12.2 in a Dart.

KB zero deck pistons is what I would do. You allready said you have the Edelbrock heads. Use as is or pocket port them for extra umph. Use with a .039 for a pump gas freindly ride. (93 octane, 9.8-1 ratio)

A good street strip tire is a BFG Drag Radials or a Mickey Tompson's ET street radial I was told did good for freind.
I think a 27 inch tire would be good.

Lose some weight with the car. S/S suspension, drag shocks etc.... will get you down to the low 12's. If you glass the cars bumpers and hoods, 11's are possible, but you'll work for them.

I would stroke it and shoot for a 10 to 1 compression with the Eddy heads. You are gaining about 70 cubic inches of engine. The 4.10 gears and 3000 stall converter will be perfect for the stroker...carb might be a bit small but if you got it run it.

Camshaft......comp HE275XL would be a nice start.

Engine will make a ton of torque....with a set of drag radials or ET street you will kill that camero.

Save the dough on stroking it. You'll beat him anyway. The 70 cubes will be in the form of torque. The carb size is fine for street, street/strip use, for all out strip, it's about 50 cfm light.

Default works 4 me

DON'T STROKE IT,with your rearend & tranny as is 340 is fine--purple shaft-509 or 484,hot ignition & BIG carb (850),must have a good stall converter (at least 3500).this combo works excellent 4 me--7.90's in the 1/8 in a 3100 pound duster--remember BIG carb,intakes are immaterial--there isn't a 10th difference in any of them--happy racing
